Jessie waited till the plantation was quiet before making a run for it. She goes down the fire ladder and hides in the bushes till night shift passes through then into the woods she goes.
She had hit her breaking point weeks before (and more rounds of the coffin) but timing had to be perfect. Or she would be sent back into the coffin by grand'mere plus being punched by TC and Jody (and much worse would happen, escapes were treated badly). So the seeds were planted as the nightgown clad woman grabs a hidden bag from under a bush, quickly changing into jeans and a sweater before throwing the gown into a tree. But a familiar glint of gold is seen around her neck as she stuffs the bag with cash, some pot she swiped from a drawer and some hockable silverware, her late mother's cross necklace (a pair of earrings are in the bag plus her father's lighter). In the process she is grazed by branches and a few hedges as she slips out of a back gate (while lighting up a cigarette) and heads for the Greyhound station and a new life. She is seen 30 minutes later on a nearby highway hitching a lift via a cover story,"My truck broke down a few miles back"...
She lands on the train after this.

Character Information
Name: Jessica "Jessie" Custer
Canon: Preacher (AMC)
Canon Point: Pre series, night she escapes Angelville and 3 months befeore she becomes a criminal with Tristan
OU/AU/CRAU/OC: AU, Rule!63
Age: 22 (best guess given timelines)
World Information: https://en.wikipedia.org/wiki/Preacher_(TV_series) (season 3 spoilers possible)
Only difference is he is now a she, looking to explore canon from a female POV.
Personal History: https://preacher.fandom.com/wiki/Jesse_Custer (season 3 spoilers possible for Angelville)
Personality: Jessica Custer is a conflicted woman. On one hand she's had enough of being the perfect robot her family wants but on the other she still wants to please her father. Her trust issues are massive since she lost Tristan (her Tulip) to the foster care system. Her rage is off the charts since fights are her idea of sport and her emotions are a mess. But there is fire, she did hatch a scheme to escape Angelville but the train had other ideas. Its the platform over the bus station but she'll take it, its a step up.
The train is freedom but can she heal not knowing what lies ahead (aka events of AMC canon). But her first step is simple: reclaiming her nickname of Jessie and her last name of Custer (under her grandmother's orders she had been Jessica L'Angelle after her parents death).
EDITS
What drives her conflicts? How does she feel about having to be a 'perfect robot' for her family and has that changed in the time she's lived with her grandma?
When she does run away Jessie has hit her breaking point, her grandmother's abuse was bad enough along with everything she took from TC and Jody. All she knew was she had to run and to not just find her soulmate but to find herself.
She has to find and embrace Jessie Custer and leave behind Jessica L'Angelle. She just has no idea where to even start which leads to a lot of her inner conflict. So she has moments of "perfect robot" and days of that true self she's having to discover anew.
The history mentioned that she prayed for her father to die and condemn him to hell- does she still hold guilt about that? Does she blame herself or others?
At the time that prayer was from a place of anger, she had just lost Tristan to foster care and had basically lost her best friend. So on one hand her head knows it was her anger kicking in and it was never her fault her heart still carries a degree of guilt even now.
How does she feel about her grandma?
Marie L'Angelle is a very touchy subject to say the least. Jessie has her theories about her somehow being a part of her mom's death but needs more to confirm her hunch. Plus she is sure years after the fact she had a role behind the death of John Custer, she just needs proof.
She is also sure she's not loved by her at all which is its own round of demons. And that's before the coffin, being locked in it underwater was a favorite tactic of the older woman to ensure perfection.
Also cue very lousy coping skills.
It only gets worse during S3.....
How does she feel about escape?
Escaping Angelville and getting her soulmate back has basically kept her sane the last few years. TC and Jody only seeing her as less than human has only made that drive stronger. Being locked in the coffin that last time only sealed it. So she kept her nose down and kept training, being the robot by day but hatching escape in private.
You mention rage, how does that manifest? What sets it off? Is she always angry? How does she tend to react/interact with people outside her family? Does she hide the anger or is she generally brash/rude etc?
Jessie is the perfect lady in public (blame grandma again) but that rage simmers under the surface. So cue lousy coping skills to keep the dark at bay from fights to chain smoking. Getting into fights keeps the worst at bay plus people learn fast not to mess with girls like her, that hook scared off more than a few (plus she has been able to use her beauty as a weapon, charming nuts out of wallets at the pool table mostly. This was learned during early test escapes before the grand running away). She also can feel things after a good brawl, she can't pinpoint why.
But she has moments of brash/rude that most assume is just a bad day or the family snark kicking in.
Is finding Tristan something she wants to do? Or does she consider him lost for good?
Finding Tristan is at the top of her post escape to do list. She knows he's out there and looking for her just the same. As kids the joke around her dad's church was they are soulmates, they really are that and more. Little does she know but she's only months from finding him.
Does she feel she is keeping her promise to her father of being 'one of the good guys'? Why? Why not? How does she feel about that?
Jessie even now tries her hardest to keep that promise from the night her dad died. She wants to keep the good of his memory alive but even now she still has moments of feeling like she can't live up to that
What are her general opinions on religion? How has being brought up by her father and then her grandma and their different extremes shaped her worldview?
John Custer may have not been the best father but he tried his best as a single father and loved his daughter completely, compared to Marie L'Angelle he was more open minded once out of her watch (like his daughter's best friend not being a "good boy"). Marie L'Angelle however is everything Jessie has vowed to never become. So Jessie is on a journey leading to what eventually happens in canon, she just has a lot of healing ahead and learning to do first. As for religion she has a lot to unravel as part of her journey and healing aka she's a work in progress.
CRAU developments: NA
Key themes: Finding her true self, escaping her past and eventual redemption
Main Motivation: Escape
Skills: Theology (divinity student and eventual preacher), expert fighter and markswoman. occult knowledge, speaks Creole, Latin and French
Item: Cigarette lighter that had been her father's and a full pack of cigarettes (she will be hunting for more)
Sample: https://nexus-crossings.dreamwidth.org/142449.html
Notes: I follow AMC canon in terms of timelines and eventual events, Tristan O'Hare is her verse's Tulip O'Hare and will CW all religious issues to be safe. She is also has severe claustrophobia thanks to being locked in a coffin who knows how many times.
I also assume jewelry is still in play since she was wearing a necklace and earrings that had been her mom's when she escaped?
